GSK Seeks Health Canada Review of Jemperli for Locally Advanced Rectal Cancer

  • GSK has submitted Jemperli (dostarlimab) for previously untreated stage II/III dMMR/MSI-H locally advanced rectal cancer, with Health Canada accepting the submission under its NOC/c policy and Project Orbis pathway.
  • The submission is supported by AZUR-1, a Phase II trial that demonstrated a meaningful and sustained clinical complete response at 12 months (cCR12), indicating no detectable cancer for at least one year after treatment completion.
  • If approved, Jemperli could potentially eliminate or delay chemotherapy, radiation and surgery for some eligible patients, potentially supporting a non-operative treatment approach.
  • Jemperli is not currently approved in Canada for this indication; its safety and efficacy in previously untreated stage II/III dMMR/MSI-H locally advanced rectal cancer remain under investigation.

Takeaways:

Approval would position Jemperli as a potential first‑line, organ‑preserving immunotherapy for dMMR/MSI‑H locally advanced rectal cancer, opening a new, biomarker‑defined segment within the ~$27B colorectal cancer therapeutics market (projected to reach ~$51B by 2034) and accelerating the shift from chemo‑radiation‑surgery toward immunotherapy‑led care. For GSK, a positive decision in Canada (alongside parallel U.S./global filings) could materially expand Jemperli’s label beyond endometrial and other tumour types, supporting mid‑to‑late‑decade oncology growth targets and creating a high‑value, guideline‑changing franchise in rectal cancer where dMMR/MSI‑H prevalence is ~5–10% of cases.
